The Stereochemistry of the Nicotinamide Adenine Dinucleotide-specific Isocitric Dehydrogenase Reaction

The stereochemistry of the agr-ketoglutarate produced in the reaction of nicotinamide adenine dinucleotide-specific isocitric dehydrogenase with threo-ds-isocitric acid-ß-T has been determined. The product retained the tritium of the substrate in the ß position without inversion. This is similar to the finding with the nicotinamide adenine dinucleotide phosphate-specific enzyme. The products of the two enzymes are therefore indistinguishable. Experiments with agr-ketoglutarate-ß-T2 provided no evidence for the reversibility of the reaction.
