Preparation and Kinetic Properties of Intermediates in the Reaction of Hemoglobin with Ligands

Eraldo Antonini 1, Maurizio Brunori 1, Jeffries Wyman 1, and Robert W. Noble 1

From the 1 From the Institute of Biological Chemistry, University of Rome; Consiglio Nazionale delle Ricerche, Center for Molecular Biology; and the Regina Elena Institute for Cancer Research, Rome, Italy

The nitric oxide derivatives of isolated alpha and ß hemoglobin chains have been prepared. By taking advantage of the very slow dissociation of NO, compounds containing the species (alphaNO - ß) or (alpha - ßNO) have been obtained by mixing NO chains with their deoxygenated partners. The kinetics of the reaction with carbon monoxide of the compounds, which may be regarded as intermediates in the reaction of hemoglobin with ligands, has been studied; it was found that both systems (alphaNO + ß) and (alpha + ßNO) behave similarly to fully unliganded hemoglobin.
