The plasmin and activator activities of highly purified streptokinase-human plasminogen and streptokinase-human plasmin mixtures of various molar ratios have been determined. The enzymatic assay data show that both human plasminogen and human plasmin participate in an equimolar stoichiometric reaction with streptokinase. The data are thus in agreement with previously reported physicochemical evidence for such a reaction. Activator activity was shown to be maximal whereas plasmin activity was minimal in equimolar mixtures of streptokinase and human plasminogen or human plasmin. The general mechanism of plasminogen activation is discussed in the light of the present findings and an hypothesis implicating the streptokinase-human plasmin complex as a universal activator of plasminogens is presented.
A streptophotometric assay procedure for the determination of plasmin is described.
