Formation of
-Ketoglutaric Semialdehyde from l-2-Keto-3-deoxyarabonic Acid and Isolation of l-2-Keto-3-deoxyarabonate Dehydratase from Pseudomonas saccharophila
-Ketoglutaric semialdehyde has been identified as an intermediate in the conversion of l-2-keto-3-deoxyarabonate to
-ketoglutarate by cell-free extracts of Pseudomonas saccharophila.
-Ketoglutarate semialdehyde arises by the intramolecular dismutation of l-2-keto-3-deoxyarabonate. The enzyme, l-2-keto-3-deoxyarabonate dehydratase, which catalyzes the conversion of l-2-keto-3-deoxyarabonate to
-ketoglutarate semialdehyde, has been purified and crystallized. The enzyme is homogeneous on starch gel electrophoresis and shows a single asymmetrical peak in the ultracentrifuge. No cofactor has been observed.
