The cover shows a structural view of the interactions of transferrin receptor (TfR) and transferrin receptor 2 (TfR2) with the hemochromatosis protein HFE and the serum iron carrier protein transferrin (Tf). TfR binds both Tf and HFE, whereas TfR2 binds only Tf. Foreground, transferrin receptor (blue ribbons) binds two molecules of HFE (red and orange ribbons) as seen in the 2.8-A crystal structure of an HFE-TfR complex. Background, HFE competes with Tf (green) for binding to TfR. HFE does not compete with Tf for binding to TfR2 (yellow; based upon the structure of TfR.
