On the cover, myosin Va and Rab27A are essential components for melanosome transport to actin-enriched cell periphery (i.e. dendrite tips; arrow), but how myosin Va recognizes melanosomes has remained undetermined. We discovered Slac2-a/melanophilin to be the "missing link" between myosin Va and GTP-Rab27A present in the melanosome. The N-terminal Slp homology domain (SHD) of Slac2-a (green) specifically binds Rab27A, and the C-terminal half (red) binds the globular tail of myosin Va.
