The cover shows the structure of the detergent-solubilized dodecameric PulD, an outer membrane secretin component of a bacterial type II secretion machinery, as determined by low dose cryo-electron microscopy. Images of particles adsorbed onto a thin carbon film and vitrified by rapid cooling in liquid ethane were mainly top views (ring-shaped), with some side views (background). The three-dimensional density map was constructed from classified projections by averaging using weighted back projection methods. The surface-rendered three-dimensional structure represents the mass of the particle as determined by scanning transmission electron microscopy. Small protrusions are thought to be the binding sites for PulS, a membrane-anchored auxiliary protein. For full details see the article by Chami et al., pages 37732–37741 .
