On The Cover: Glucosidase II plays a critical role in glycoprotein processing and quality control in endoplasmic reticulum. Structurally defined high mannose-type glycans were synthesized systematically and converted to their protein conjugates. These molecules revealed the substrate specificity of glucosidase II, both in the presence and absence of lectin chaperone calreticulin.
