On The Cover: Many people are suffering from allergies (e.g. anaphylaxis, cedar pollen). The symptoms are triggered by cross-linking of the Fc receptor, FcεRI, on mast cells with IgE bound to allergens. As such, IgE is a key target for allergic therapy. Here, we demonstrate that the commercial Vibrio cholerae culture fluid (receptor-destroying enzyme (RDE) (II)) can specifically inactivate IgE and not IgG. RDE (II) was found to modulate the structure of IgE, reducing the level of binding to antigen and FcεRI on mast cells, thereby abrogating anaphylaxis. The main role of RDE (II) was found to be in its influence on the branched glycan of IgE. This study provides a potential new candidate for therapy against allergic diseases.
