On The Cover: Antigen-binding fragment (Fab) of an antibody depicted as a sphere model (cyan) is engineered to bind two structurally unrelated angiogenic factors, vascular endothelial growth factor (orange) and angiopoietin 2 (pink). This dual action Fab achieved sub-nanomolar affinity for both antigens, a feat enabled by deep mutational scanning, which measured the effect of all possible mutations of each CDR residue depicted by heat map (left background) and identified synergistic mutation pairs by cooperativity analysis represented with the Circos plot (right background).
